Mezopotamia leży w dorzeczu dwóch wielkich rzek: Eufratu i Tygrysu. Jej obszar (często różnie pojmowany, jeśli chodzi o zasięg) rozciąga się od północnej Syrii aż po Zatokę Perską i od Iranu aż po Arabię. W innym, bardziej precyzyjnym ujęciu, można Mezopotamię zamknąć w trójkącie: Aleppo – jezioro Urmia na północy i ujście rzeki Szatt al-Arab (rzeka powstała po połączeniu Eufratu i Tygrysu) do Zatoki Perskiej na południu.
Granice geograficzne Mezopotamii przedstawiają się następująco: od strony północnej jest ona ograniczona łańcuchami górskimi południowo-wschodniej Anatolii (m.in. Taurus), które stopniowo łączą się ze wschodnią granicą Mezopotamii, którą jest potężny łańcuch górski Zagrosu, oddzielający ją od Wyżyny Irańskiej. Od strony południowej, na dość krótkim odcinku, granicą Mezopotamii jest linia brzegowa Zatoki Perskiej, a następnie, dochodząc do pustyni Wielki Nefud i Syryjskiej, granica Mezopotamii biegnie równolegle do biegu Eufratu wzdłuż wymienionej pustyni w kierunku północnym aż do północnej Syrii.
